Just enter the actual level 7 via the stairs, then go back to the fighters challenge, that's all that's required. Level 7 proper, not falling down a pit or any other access points... the stairway through Sequencis that you need the round keys to get to.
Don't know why it would be stuck, one of the first things I did hitting floor 6 on my second playthrough was access this. As soon as I visited 7 proper and came back, I had access.

Just went through on a fresh save (just got to level 6). The door opened after I had been to level 7 and had the Sword of Nex. I didn't do anything special or uncover any other secrets.

Well I got access without entering lvl 7 properly, See my previous post to what Was the key to open the gate in my case(I had plenty of other gear on my to like the orb etc, but since it states "fighters challange" and it was the actual sword that opend the door, I'm fairly certain that is the what is required and not all the loot from the whole lvl.)
Someone in your party need to carry Sword of Nex, not necessarily equipped

Where is that sword located ?
It is in the "pit room" where the fire dudes spawn. you get it when u complete the button/pit fall puzzle in that room.
And yes as the above poster states, sword of flame might be a req to, I had it on me for the most part of the level so I didn't really think about it. But that sword shouldn't be an issue since u get it before u get to the "fighters challenge" anyway if memory serves me right.
I can confirm it is ONLY about Sword of Nex (Sword of Flames is not required).
After finishing lvl 10 i decided to go back to Fighter's Challange. But it didnt open.
I read this topic and few people stated Sword of Nex is needed (which i DIDNT had in possesion by the time of trying to reach FC door). So i backtracked few lvls in search of that sword (got better and dumped Nex one), somehow i found it (got a habbit to toss items inside Iron Door/near SaveCrystals. When i got it back and returned to FC door it opened.
So - only Sword of Nex looks like required (by that time i had 13/14 lvl chars and 35+sword/axe skills).
Managed to open it today without taking a trip down to level 7 (other than falling into pits - maybe it counts). I had completed all the level except the last round key and had the Sword of Nex.

Depends on your team setup really but on hard difficulty, I couldn't prevent my mage from dying in the dozen or so attempt I gave it, so I gave up and just finished it with 3. I tried equipping platearmor on him and casting invis before going in and using pots but his 60life just wasn't cutting it so I just used him to insta kill 2mobs then let him die.
Trick is rush straight ahead when you go in until you reach the wall, then pick a side(generally the side on which your mage is since he's dead) and go straight until you reach the corner. All the while killing whatever is on your way and spamming forward so you don't get blocked. And make a bunch of healing potions beforehand and use them until you get in the corner. Once there, keep your best tank(preferably evasion) on the room side so he's the only one taking damage from the side mob, and make sure to remake potions/drink them with him so he doesn't die while you kill everything. I also popped a potion of speed on my archer so he'd shoot more often since he does the most damage per round with double arrows, and I switched to crossbow when I ran out of arrows.
On normal it's a lot more forgiving so you don't need quite as much of an optimized setup, but I feel it's a good gauge of how strong your party is. Can always come back later(not too much later but still you could get up to lvl 10-11 easily before coming back which would make it easier).
As a side note, in one of my worst attempt, I only got my evasion rogue to reach the corner. I still killed the rest of the room solo. Evasion really is balanced ^^.
